The Wolen and Gruber method required two extractions into butyl chloride, a freezing step to break the emulsion, an extraction into an aqueous acid solution, back extraction into chloroform at pH 11, and concentration of the extract before GLC analysis. No interference studies were conducted. In the method of Manno et al. 10 ; , there are two extractions into an organic phase, an extraction into an acid layer, concentration of the extract under reduced pressure, and a rinse step, followed by drying of the extract under nitrogen. They did not assess interference, recovery, or precision. In summary, all methods to date have been lengthy and time-consuming, require multiple sample-preparation steps, often lack specificity, and are inadequately examined for accuracy and precision. We present two isothermal GLC methods for prkpoxyphene itself, which require only a single extraction and no derivative formation. One of these methods is sensitive enough for measuring concentrations corresponding to those in serum after the usual therapeutic dosage "therapeutic method" the other is designed for toxic concentrations "toxic method.
