MA treatment also produced an impairment in the Morris water-maze place task at 65 days postinjection. No impairments were observed in water-maze moving platform or place task at 79 and 165 days postinjection, respectively, or in T-maze alternation. Since evidence suggests that melatonin may act as a free radical scavenger and antioxidant, the present study was undertaken to investigate the effect of melatonin on METHand MPTP-induced neurotoxicity. In addition, the effect of melatonin on METH-induced locomotor sensitization was investigated. The administration of METH 5 mg kg -1 ; x 3 ; or MPTP 20 mg kg -1 ; x 3 ; to Swiss Webster mice resulted in 45-57% depletion in the content of striatal dopamine and its metabolites, 3, 4-dihydroxyphenylacetic acid and homovanillic acid, and 57-59% depletion in dopamine transporter binding sites. The administration of melatonin 10 mg kg -1 before each of the three injections of the neurotoxic agents on day 1 ; , and thereafter for two additional days, afforded a full protection against METH-induced depletion of dopamine and its metabolites and dopamine transporter binding sites. In addition, melatonin significantly diminished METH-induced hyperthermia. However, the treatment with melatonin had no significant effect on MPTP-induced depletion of the dopaminergic markers tested. In the set of behavioral experiments, we found that the administration of 1 mg kg -1 ; METH to Swiss Webster mice for 5 days resulted in marked locomotor sensitization to a subsequent challenge injection of METH, as well as context-dependent sensitization conditioning ; . The pretreatment with melatonin 10 mg kg -1 prevented neither the sensitized response to METH nor the development of conditioned locomotion. Results of the present study indicate that melatonin has a differential effect on the dopaminergic neurotoxicity produced by METH and MPTP. Since it is postulated that METH-induced hyperthermia is related to its neurotoxic effect, while regulation of body temperature is unrelated to MPTP-induced neurotoxicity or METH-induced locomotor sensitization, the protective effect of melatonin observed in the present study may be due primarily to diminishing METH-induced hyperthermia. 267 UI - 9696171 AU - Richards JR AU - Derlet RW and metaproterenol.
Exposure to residential magnetic fields may disrupt the normal nocturnal rise in melatonin levels, resulting in increased risk for breast cancer, possibly through increased levels of reproductive hormones. The authors investigated whether exposure to a 60-Hz magnetic field under controlled conditions is associated with a decrease in urinary nocturnal 6-sulfatoxymelatonin level and increase in luteinizing hormone LH ; , follicle-stimulating hormone FSH ; , and estrogen levels in healthy premenopausal women. Using a crossover design, half the participants were assigned to magnetic field exposure of 5 to greater than ambient levels for 5 consecutive nights during the early to midluteal phase of the menstrual cycle. On the last night of exposure, a nocturnal urine sample was collected. The next month, participants were sham exposed. The other half of participants were assigned the reverse order of exposure. Magnetic field exposure was associated with decreased 6-sulfatoxymelatonin levels, but no changes in reproductive hormone levels were observed. Participants using prescription medications and anovulatory participants had more pronounced decreases in 6-sulfatoxymelatonin levels with magnetic field exposure. Conclusion: This study provides further evidence that exposure to magnetic fields is associated with decreased nocturnal melatonin levels, but does not support the hypothesis that such exposure results in increased urinary levels of estrogens, LH, or FSH.

With a growing awareness of the potential hazards of chemical and biological agents in our homes, schools and workplaces, a greater emphasis has been placed on evaluating the role of mycotoxins and mycotoxin-producing fungi in our indoor environments. Now, more then ever, the potential health effects of airborne molds and other biological contaminants are being given serious consideration in IAQ investigations. What are mycotoxins? Simply, a mycotoxin is any toxic fungal metabolite. Fungi produce a variety of secondary metabolites as a byproduct of their metabolism. Those capable of eliciting deleterious effects on other organisms are classified as mycotoxins. Mycotoxicosis can be defined as illness resulting from ingestion, inhalation, or other involvement with mycotoxins. Mold mycotoxins of primary concern here elicit a harmful effect on humans. Other substances such as those produced by poisonous mushrooms are also mycotoxins, and mycetismus is the term applied to mushroom poisoning as a distinct category of mycotoxicosis. The discovery of antibiotics produced by fungi revolutionized the treatment of disease because the compounds are sufficiently non-toxic to the host. In nature, mycotoxins are a chemical defense for fungi and have evolved as mechanisms for antiherbivory and to provide a competitive advantage when colonizing new substrata. Historical Mycotoxicoses The study of mycotoxins originally centered on the health effects of ingestion of toxic fungal byproducts from growth of fungi on food and livestock feed. A review of the historical significance of mycotoxins sets the stage for more recent work with inhalation myctoxicosis studies and reglan.

Receptors, 73, 79, 9597 but it has been demonstrated98, 99 that glutathione-S-transferases are the predominant LTC4 binding site in many tissues, masking LTC4 binding to its true receptors. Recently, in HLP membranes we have demonstrated the presence of a high affinity binding site Kd 0.015 nM ; , specific for 3H-LTC4 and endowed with receptor characteristics, but lacking sensitivity to GTP.66 To date, there is no clear evidence for this site to be a functional LTC4 receptor while its pharmacological characterization has been undertaken100. CysLT2 receptors have been shown to be present, together with CysLT1, in GP lung6870 and trachea71, 72 and rat lung.73 The binding data in this latter tissue73 are at variance with functional data, which suggest the presence of a homogeneous population of CysLT1 receptor.64, 74 The presence of a CysLT2 receptor in human bronchi is debated, because BAYu9773 does indeed inhibit LTD4-induced bronchoconstriction in vitro, but its potency is quite low, as already mentioned.63 In contrast, a homogeneous population of CysLT2 receptor has been identified in ferret bronchus101 and spleen, 102 sheep bronchus and trachea 103 and human pulmonary veins.63 Very recently, the CysLT2 receptor has been cloned and has been shown to couple to [Ca2 + ]i elevation in Xenopus oocytes and HEK 293 cells, displaying equal sensitivity to LTC4 and LTD.450 These increases are not affected by CysLT1 classical antagonists. Large amounts of the CysLT2 mRNA have been found in lung macrophages and airway smooth muscle, human heart, adrenal medulla cells, peripherial blood leukocytes and brain, 50 and by other authors in placenta and spleen.51 The receptor gene has been mapped to chromosome 13q14, a region linked to atopic asthma.50, for example, melatonim alcohol. In the summary judgment application of Calgon Carbon Corporation v. The RAKFKQLLQ specificity of these colonies was confirmed by the rapid T cellT cell assay and 51Cr-release assays data not shown ; . In all colonies except one [BT III ; 36] a single in-frame TCR rearrangement was detected Fig. 6A ; . Each time point analysis revealed a unique set of TCR responding to RAKFKQLLQ, with no patterns of TCRV, TCRJ or CDR3 amino acid conservation. The only structurally similar clones were BT I ; 74 Fig. 5 ; and BT II ; 114 Fig. 6A ; that shared BV6 gene usage and the CDR3 amino acid sequence, SLILXG. Interestingly, the CDR3 loop length restriction that was evident during primary infection continued to be selected for in persistent infection, with all the colonies having a CDR3 length of 811 amino acids. Altogether, this longitudinal analysis showed no dramatic RAKFKQLLQ clonotype selection, but rather the maintenance of TCR diversity. TCR chain sequence analysis was also extended to an HLA B8 type A and type B healthy virus carrier, ISM and MS respectively Fig. 6B ; . The RAKFKQLLQ specificity of these colonies was confirmed by the rapid T cellT cell assay and 51Cr-release assays data not shown ; . As was the case in primary infection, non-recurrent rearrangements with a diverse TCR repertoire usage and a restricted CDR3. Synopsis According to a report published in the July issue of the journal Heart, patients with atrial fibrillation AF ; are nearly 20 times more likely to be infected with the common gastric microbe Helicobacter pylori than are healthy controls, with the association strongest in patients with persistent rather than paroxysmal atrial fibrillation. AF patients also had higher levels of C-reactive protein CRP ; than did controls, suggesting that H. pylori's effects on this arrhythmia may be mediated through an inflammatory pathway. The researchers carried out a study investigating H. pylori positivity and CRP levels in 59 patients with AF and 45 healthy controls. The AF group was further divided into 29 patients with persistent disease and 30 with paroxysmal disease. In the overall analysis, 97.2% of AF patients were seropositive for H. pylori compared with just 5.3% of controls. As noted, CRP levels were significantly higher among AF patients as well p 0.001 ; . Further analysis showed that persistent AF was tied to a higher rate of H. pylori seropositivity p 0.027 ; and to higher CRP levels than was paroxysmal disease p 0.041 ; . Although the exact mechanism is unknown, the researchers hypothesise that it may relate to autoantibodies that develop in some H. pylori-infected patients. These antibodies, which normally attack a proton pump found on gastric cells, may instead attack a similarly appearing pump on cardiac cells, ultimately triggering AF. The researchers conclude that their findings show a significant link between H. pylori seropositivity and AF. However they add, "More data will be necessary from controlled studies to further identify how H. pylori can influence the pathogenesis of AF".
